Abstract
In The Lancet Neurology, Pierre N Tariot and colleagues present the Alzheimer's Prevention Initiative (API) Colombia Trial, a landmark public–private partnership, evaluating crenezumab in autosomal-dominant Alzheimer's disease (ADAD). The API ADAD Colombia Trial enrolled 252 members of the world's largest kindred in Medellín, Colombia, all carrying the PSEN1 Glu280Ala mutation, a population whose well characterised natural history, defined through decades of work by the late Francisco Lopera and colleagues, made it uniquely suited for a preventive intervention.
